Toxicity and resistance to chemotherapy drugs remain major hurdles in oncology. My proposal is focused on using an emerging genetic strategy called "haploid genetic screening" to discover genes that mediate resistance to topoisomerase inhibitors, chemotherapeutics which form the backbone of treatment for several cancers.

Background

Traditional approach targeting only genetic factors and non-cancer stem cells for drug discovery appears insufficient to combat cancer diseases especially in the category of childhood leukemia caused by MLL gene mutation which occurs frequently in infants and children and associates with poor survival rate. It is becoming clear that cancer cells transformation is initiated and maintained by both genetic and epigenetic mechanisms.

Acute lymphoblastic leukemia (ALL) is the most common malignancy in children. Although great strides have been made in improving outcomes for children with ALL, some groups of children will not survive this disease. Recent discoveries have found that genetic mutations in proteins that are involved in normal lymphocyte development play a role in the development of leukemia, including a protein called Ikaros. The mechanism by which Ikaros causes leukemia is unknown.

Project Background

Neuroblastoma is one of the most common cancers in children, only leukemias and brain cancers are more frequent. The five-year survival for children with metastatic neuroblastoma is only 35%. With recurrent disease, this percentage is even lower. In addition, all modalities currently used in treating cancers including chemotherapy, radiation, and surgery are utilized in these patients. For a four year old, this can encompass a large portion of his or her short life. This is a disease that medicine must have greater success with in the future.
