A significant portion of Ms. Stiene's project will consist of characterizing Ewing Sarcoma (ES) cell line migration and metastasis in our zebrafish ES xenograft model using ImageJ software. Our lab has created a ES xenograft model in zebrafish by injecting RFP-labeled ES cell lines into the transgenic line Tg(fli:GFP). This line marks the zebrafish vasculature green, allowing for effective tracking of the injected red cancer cells. To optimize our xenograft model, we are methodically characterizing tumor¬ invasion and metastatic dissemination of the labeled ES cell lines within the zebrafish. Labeled cells are injected at 2 days post fertilization (dpf) and cell growth and migration are tracked until 5 dpf. Using fluorescent microscopy, images of each injected fish are collected at one day post injection (corresponding to 3 dpf) and three days post injection (corresponding to 5 dpf). Using ImageJ software, these images are carefully analyzed for tumor growth, cell migration, and metastasis.
